基于DNA条形码(ITS2)的中药材防风与其混伪品的鉴定

[摘要] 目的 通过ITS2序列分析,对防风及其易混伪品进行鉴定,确保用药安全。 方法 从GenBank数据库下载中药材防风与其混伪品的DNA条形码ITS2序列,利用MEGA 6.06软件进行比对分析,计算GC含量及变异位点、种内、种间遗传距离,并构建NJ系统发育树,应用ITS2数据库网站预测其ITS2二级结构。 结果 序列分析与遗传距离结果显示,防风与其混伪品存在明显差异,基于ITS2序列的NJ树及其ITS2二级结构均可以将防风与其混伪品进行区分。 结论 ITS2能够准确鉴定防风及其易混伪品,可用于中药材的快速鉴别。

[Abstract] Objective To identify Saposhnikovia divaricata and its adulterants by using ITS2 sequence analysis and to secure medication safety. Methods ITS2 sequences of Saposhnikovia divaricata and its adulterants were downloaded from GenBank,MEGA 6.06 software was used for sequence analysis to summarize GC content,variation sites,estimate intra-and inter-specific genetic distances,and built NJ trees.Moreover,the secondary structure of ITS2 was predicted by using the ITS2 database websites. Results ITS2 sequence analysis and genetic distances calculation showed significant difference in the samples.Further more,NJ tree and secondary structure results could distinctly differentiate quality product and adulterants. Conclusion ITS2 can effectively identify Saposhnikovia divaricata and its adulterants,as a result,it can be used for rapid identification of Chinese medicinal materials.

防风(Saposhnikovia divaricata)是双子叶植物伞形科(Umbelliferae)多年生草本植物防风属(Saposhnikovia)防风的干燥根[1],是常用的中药材之一,主要产于我国北部和东北部,分布于东北、华北及陕西、宁夏、甘肃、山东等地[2]。防风具有祛风解表、除湿止痛、疏肝解痉、杀虫止痒的功效,主治外感风寒,头痛项强,目眩昏涩,风寒湿痹,骨节酸痛,腹痛泄泻,肠风下血,破伤中风,麻疹难透,风疹瘙痒,疮疡初起。由于防风的药用价值高,市场需求极大。近年来,部分不法商人以川防风、杏叶防风、华中前胡、田葛缕子、硬阿魏、野萝卜等冒充正品防风,影响了用药疗效及安全。传统的防风鉴定方法主要是从基原、性状、显微、理化等方面进行区别[3-5],这些方法对操作人员的专业知识要求较高,具有一定的局限性,难以满足中医药现代化发展的要求,因此,亟需建立一种简单、快速、准确的鉴定防风药材及其混伪品的方法。
DNA条形码[6](DNA barcoding)技术是用一段标准的、较短的DNA序列作为物种标记的一种物种分子鉴定方法,可以对生物物种实现准确、快速、简便的鉴定。“DNA条形码”首先由加拿大动物学家、皇家学会会员Paul Hebert于2003年提出,是在分子分类学和DNA分类学的基础上用单一的小片段基因作为物种的条形码,进而为整个生物界的物种编码,达到类似超市利用商品条形码可以快速准确识别成千上万种商品的效果[7]。
